Captain Barbarella, a space pilot from Earth, has been given a mission from the President of Earth to find a rogue pilot named Durand Durand who disappeared in the Tao Setti system. Durand Durand is a scientist who invented the positronic ray, a weapon of mass destruction.

(for the movie purists. I have used creative license here.The fembot Mado however, is canon. The original comics in the 1960's had Mado as a canon character, Barbarella's faithful companion who always wanted to be more human.)
